چکیده

UNLABELLED PREMISE OF THE STUDY Microsatellite markers were developed to assess the level of genetic variation and population structure in Banksia integrifolia, a widespread species endemic to eastern Australia. • METHODS AND RESULTS We used next-generation sequencing approaches to identify and develop 11 polymorphic microsatellite markers with perfect tri- and tetranucleotide repeats. We tested these markers with 71 specimens from three populations. Observed and expected heterozygosities ranged from 0.0 to 0.875 and 0.0 to 0.763, respectively. • CONCLUSIONS The developed markers will be valuable for studies of the population structure, mating system, and selection of provenances for restoration projects involving B. integrifolia.

Whole genome shotgun sequences for microsatellite discovery and application in cultivated and wild Macadamia (Proteaceae)1

UNLABELLED PREMISE OF THE STUDY Next-generation sequencing (NGS) data are widely used for single-nucleotide polymorphism discovery and genetic marker development in species with limited available genome information. We developed microsatellite primers for the Proteaceae nut crop species Macadamia integrifolia and assessed cross-species transferability in all congeners to investigate genetic ...

First instalment in resolution of the Banksia spinulosa complex (Proteaceae): B. neoanglica, a new species supported by phenetic analysis, ecology and geography

Taxa in the Banksia spinulosa Sm. complex (Proteaceae) have populations with sympatric, parapatric and allopatric distributions and unclear or disputed boundaries. Our hypothesis is that under biological, phenetic and diagnosable species concepts that each of the currently named taxa within the Banksia spinulosa complex is a separate species. Based on specimens collected as part of this study, ...

Species of Botryosphaeriaceae occurring on Proteaceae

The Botryosphaeriaceae includes several species that are serious canker and leaf pathogens of Proteaceae. In the present study, sequence data for the ITS nrDNA region were used in conjunction with morphological observations to resolve the taxonomy of species of Botryosphaeriaceae associated with Proteaceae. Neofusicoccum luteum was confirmed from Buckinghamia and Banksia in Australia, and on Pr...

Estimating seed bank accumulation and dynamics in three obligate-seeder Proteaceae species

The seed bank dynamics of the three co-occurring obligate-seeder (i.e. fire-sensitive) Proteaceae species, Banksia ericifolia, Banksia marginata and Petrophile pulchella, were examined at sites of varying time since the most recent fire (i.e. plant age) in the Sydney region. Significant variation among species was found in the number of cones produced, the position of the cones within the canop...

Luteocirrhus shearii gen. sp. nov. (Diaporthales, Cryphonectriaceae) pathogenic to Proteaceae in the South Western Australian Floristic Region

Morphological and DNA sequence characteristics of a pathogenic fungus isolated from branch cankers in Proteaceae of the South West Australian Floristic Region elucidated a new genus and species within Cryphonectriaceae (Diaporthales). The pathogen has been isolated from canker lesions in several Banksia species and Lambertia echinata subsp. citrina, and is associated with a serious decline of t...
